- 以细叶百合为试材,采用组织培养的方法,研究细叶百合不同部位、不同激素配比及不同发生途径对获得细叶百合再生植株的影响。结果表明:以鳞茎为外植体,通过器官发生途径获得再生植株,其最佳分化及增殖培养基为MS+6-BA 1.0 mg/L+NAA 0.1 mg/L;以花器官、幼嫩叶片为外植体通过体细胞发生途径获得再生植株,其愈伤组织诱导培养基为MS+2,4-D 2.0 mg/L+NAA 1.5 mg/L,不定芽诱导培养基为MS+6-BA 1.0 mg/L+IAA 0.1 mg/L,最佳增殖培养基为MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,生根培养基为1/2MS+NAA 0.1 mg/L。
- Abstract:
- Taking Lilium pumilum as experimental material,using tissue culture method,effects of different parts of Lilium pumilum, different hormone combinations and different ways of plant regeneration on Lilium pumilum were studied.The results showed that with the bulb as explants,the organ regeneration plants were obtained,the best medium for differentiation and proliferation was MS+6-BA 1.0 mg/L+NAA 0.1 mg/L;in flower organ,the young leaves as explants via somatic embryo genesis regeneration plants were obtained,and the callus induction medium was MS+2,4-D 2.0 mg/L+NAA 1.5 mg/L,adventitious bud induction medium was MS+6-BA 1.0 mg/L+IAA 0.1 mg/L,the optimal proliferation medium was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L.Rooting culture medium was 1/2MS+NAA 0.1 mg/L.
