- 以黑果腺肋花楸带芽茎段为试验材料,采用组织培养的方法,研究不同激素含量和配比对其组织培养和快速繁殖的影响。结果表明:茎段在MS+6-BA 0.5 mg/L+NAA 0.2 mg/L培养基上诱导芽最快最好,KT能诱导愈伤组织,但对试管苗的芽诱导无明显影响;在MS+6-BA 0.5 mg/L+IBA 0.2 mg/L培养基上继代1次,增殖系数就能达6倍以上;生根培养基1/2MS+IBA 0.5 mg/L效果最好,生根率近100%;移栽基质以草炭土∶营养土∶蛭石=1∶2∶1的基质中最好,成活率达100%。
- Abstract:
- Taking Aronia melanocarpa in vitro as the experiment materias,and the optimal tissue culture and rapid propagation system were studied by using the method of tissue culture,the effects of different hormone contents and proportion on the tissue cuiture rapid propagation was researched.The results showed that MS+6-BA 0.5 mg/L+NAA 0.2 mg/L was the best culture medium to induce budlet from its tuber,KT could induce callus,but it had no effect on test-tube plantlets’buds inducing.MS+6-BA 0.5 mg/L+IBA 0.2 mg/L was the best culture medium to induce proliferation buds from its tuber,and the propagation coeficient was six times or above.The best culture medium for multiplication of cluster shoots was 1/2MS+IBA 0.5 mg/L,the rooting rate achieved 100%.The best transplanted matrix was turfy soil∶nutrition soil∶vermiculite=1∶2∶1,and the survival rate approximated 100%.
