- 为了筛选铁皮石斛优良种质,在广西、安徽、云南、浙江收集不同产地的铁皮石斛,参照2010年版《中国药典》提供的方法,采用苯酚-硫酸法测定多糖含量,柱前衍生化HPLC法测定甘露糖含量,并使用高效液相法测定毛兰素的含量。结果表明:在0.018~0.090 mg范围内,葡萄糖与吸光值线性关系良好,相关系数r=0.9987,不同产地铁皮石斛多糖含量为 25.15%~36.53%;柱前衍生化HPLC法测定D-甘露糖含量专属性强、精确、重现性好,不同产地铁皮石斛甘露糖含量为18.00%~32.35%;不同产地铁皮石斛毛兰素的含量为0~0.0043%。不同产地的铁皮石斛多糖、甘露糖和毛兰素含量差异显著,其中采自云南、浙江的种质在多糖和甘露糖方面有优良的遗传基础,而广西种质的毛兰素含量相对较高。
- Abstract:
- To screen the elite germplasm of Dendrobium officinale,the content of polysaccharides,D-mannose and erianin from different habits were studied in this paper.According to the methods from the Pharmacopeia of the People’s Repubic of China,the content of polysaccharides was determined by phenol-vitriol colorimetric,and using Pre-column derivatization-HPLC measured D-mannose content,and the content of erianin determined by HPLC.The results showed that a good correlation (r = 0.9987) between glucose level and absorbance values at range from 0.018 to 0.090 mg.The content of polysaccharides of Dendrobium officinale ranged from 25.15% to 36.53%.Pre-column derivatization-HPLC method for the determination of D-mannose content was specific,accurate,reproducible.The D-mannose content of Dendrobium officinale ranged from 18.00% to 32.25%.The contents of erianin ranged from 0 to 0.0043%.There were singnificantly difference in the content of polysaccharides,D-mannose and erianin of Dendrobium officinale from different habits.
