- 选取“东方蜜一号”甜瓜的新生叶片为外植体,接种于含有不同激素配比的MS培养基中,诱导愈伤组织、不定芽与不定根的形成,比较不同激素浓度对外植体脱分化和再分化的影响。结果表明:“东方蜜一号”甜瓜叶片离体培养的最佳愈伤组织诱导培养基为MS+2,4-D0.10 mg/L+6-BA 1.50 mg/L,诱导率达91%,最佳不定芽诱导培养基为MS+2,4-D 0.01 mg/L+6-BA 1.50 mg/L,平均不定芽个数为3.52个;最适生根诱导培养基为MS+2,4-D 0.05 mg/L+NAA 0.20 mg/L+IBA 0.60 mg/L,生根率达85%。
- Abstract:
- New borning leaves of ‘Dongfangmi No.1’ melon were selected as explant and planted into the MS medium with different concentration and variety of endocrine to induce the formation of callus and adventitious buds and roots.The results showed that the most ideal medium for callus inducing was MS+2,4-D 0.10 mg/L+6-BA 1.50 mg/L with the best induction rate 91%;The most ideal medium for adventitious buds inducing was MS+2,4-D 0.01 mg/L+6-BA 1.50 mg/L with the best average 3.52;the best rooting medium was MS+2,4-D 0.05 mg/L+NAA 0.20 mg/L+IBA 0.60 mg/L with the best rooting rate 85%.
