- 利用特异引物对3份抗病纯合材料(基因型Ty-1/Ty-1)和4份感病纯合材料(基因型ty-1/ty-1)进行PCR扩增,抗、感材料均产生398 bp的PCR扩增片段,抗病和杂合抗病材料的酶切产物存在Taq I 酶切位点,酶切后分别产生了303和98 bp以及398、303 和98 bp的片段,而纯合感病材料的扩增产物无此酶切位点,酶切后仍为398 bp的片段。该标记能够区分抗病材料、感病材料及抗病杂合材料,是与番茄黄化曲叶病毒病抗病基因Ty-1紧密连锁的共显性标记。利用该标记对13份番茄杂交种进行检测,有8份杂交种含有Ty-1抗病基因,3份材料不含Ty-1抗病基因。利用这条标记对国内的13份番茄自交系进行检测,13份材料均不含Ty-1抗病基因。
- Abstract:
- Three resistant homozygous lines (Ty-1/Ty-1) and four homozygous susceptible lines (ty-1/ty-1) were amplified by specific primer,both resistant and susceptible lines produced PCR product about 398 bp,after digestion with enzyme Taq I,resistant genotypes and heterozygous resiatant genetypes could produce fragment about 303 bp,98 bp and 398 bp,303 bp and 98 bp respectively.Susceptible genotypes still presented 398 bp fragment.This marker could distinguish resistant and susceptible lines,was a co-dominant marker tightly linked to Ty-1 gene.13 hybrids were detected using this marker,and the results showed that 8 hybrids confer Ty-1 resistant gene and 3 hybrids do not confer Ty-1 resistant gene.The marker also were used to detect 13 inbred lines and all of them do not confer Ty-1 gene.
