辣椒疫霉菌复壮与保存及生长特性研究

- 摘要:
- 研究了辣椒疫霉菌3个生理小种的孢子囊诱导、回接复壮、短期固体培养基保存方法和复壮、培养基厚度、菌龄、生理小种对菌落生长及其形态的影响。结果表明:打菌饼诱孢较整皿光照诱孢速度快、产孢量大;发病辣椒茎段较发病叶片回接效率高,达到50%;疫霉菌在16℃下固体培养基上保存效果最好,保存时间为3~4个月;辣椒疫霉菌复壮前后Ph1、Ph3生理小种菌丝生长速率无显著差异,Ph2有显著差异,复壮后3个生理小种菌丝都变致密,致病性增强;培养基厚度可影响菌丝生长速率,10~15mL体积的培养基较适合疫霉菌的培养;疫霉菌菌丝的菌龄对菌丝生长速率无影响;Ph1、Ph2、Ph3随着致病性的增强菌丝生长速率下降。

- Researchesonmethodsofsporangiuminducing,rejuvenation,preservationandgrowthcharacteristicsofPhytophthoracapsiciwerecarriedout.Theresultsshowedthatthesporangiumonmycelialplugsweremoreandappearedearlierthanthemyceliumpickingoutfromthewholecolonyunderlight;Theefficiencyofstemsegments’backinoculationwashigherthanthatoftheleafsegments;ThepreservationeffectonPSAmediumunder16℃wasbestandpreservationtimewasthreetofourmonths;GrowthrateofPh1andPh3strainsexceptPh2hadnosignificantdifferencespre-andpost-rejuvenationbutthecolonyofthreephysiologicalracesbecamecompactandpathogenicityenhancedafterrejuvenation;Thethicknessofmediumhadeffectonthegrowthofcolonyandthebestvolumewas10~15mL;Thecellageofmyceliumhadnoeffectonthegrowthofcolony;Thegrowthrateofthreephysiologicalracesslowedastheirpathogenicityenhanced.
