核桃内果皮RNA提取方法研究

- 以核桃果壳硬化期的内果皮为试材,针对核桃内果皮木质化程度高且富含多酚多糖的特点,比较CTAB法Ⅰ、CTAB法Ⅱ和试剂盒3种方法提取核桃内果皮RNA的效果。结果表明:3种方法提取的RNA均可见3条带。CTAB法Ⅰ提取的RNA略微拖带,OD-260/OD280为1.540,RNA产率为75.9μg/g,RNA完整性差,质量低;CTAB法Ⅱ的RNA图谱较完整,但亮度较弱,OD-260/OD-280为1.801,产率为86.1μg/g,但RNA提取过程耗时太长;试剂盒法提取的RNAOD-260/OD-280为2.055,RNA产率96.4μg/g,RNA图谱清晰、稳定、明亮,证明得到的RNA多糖及酚类等去除较彻底,纯度、完整性和产率均较高。因此,应用复杂植物总RNA提取试剂的试剂盒法可获得高质量与高产率的核桃内果皮总RNA,完全满足后续分子生物学研究。

- JuglansregiaL.endocarpwereemployedasmaterialsinthisreport,becauseJuglansregiaL.endocarphadhighdegreeoflignificationandendocarpwererichinpolysaccharidesandpolyphenolcompounds,theperformancesofthreeextractionmethodsoftotalRNA:CTAB1,CTAB2andRNAplantReagentwerecomparedusingintheJuglansregiaL.endocarp.TheresultsshowedthatthetotalRNAisolatedbythreemethodshadthreebands.ThetotalRNAisolatedbythemethodofCTAB1hadtrailedbands,theOD-260/OD-280valueoftotalRNAwas1.540,theyieldwas75.9μg/g,theRNAintegralityandqualitywerefaulty.ThetotalRNAisolatedbythemethodofCTAB2hadintactbands,butitwascaliginous,theOD-260/OD-280valueoftotalRNAwas1.801,theyieldwas86.1μg/g,butthetimeofRNAisolationwasconsumed.TheOD-260/OD280valueofRNAisolatedbythemethodofRNAplantReagentwas2.055,theyieldwas96.4μg/g,threebandswereclean,steadyandbright,thatprovedthemethodcouldremovepolysaccharidesandpolyphenolcompoundscompletely,itcouldextracthighpurityandcompleteRNAatahighproductivity.ThetotalRNAisolatedbythemethodofRNAplantReagentwashighqualityandproductivity,themethodcompletelywasfitforfurtherbiologicalresearch.
