月季病原真菌分离鉴定及拮抗菌筛选

- 以南阳发病月季茎叶为试材,采用组织分离法分离病原真菌,并进行分子生物学鉴定;通过平板对峙法筛选拮抗菌,并进行离体致病性测定,确定引起病害的真菌种类并进行拮抗菌筛选,以期为月季早期真菌病害的微生物防治提供参考依据。结果表明:基于形态学、ITS序列分析B8为Colletotrichum aenigma,B14为Botryotinia ranunculi,B39为Nigrospora musae,B44为Fusarium longifundum;致病性测定试验发现4株真菌均为月季病原真菌;细菌拮抗试验发现贝提卡假单胞菌、诺丽鞘氨醇单胞菌和皮尔瑞俄类芽孢杆菌对4种病原菌的抑菌率均达到90%以上;真菌拮抗试验发现巨黑木霉、黑褐木霉、非洲哈茨木霉和云南木霉对4种病原菌的抑制效果不尽相同,对B8、B39和B44均有一定的抑制作用,而对B14的抑制效果均不太理想。综上,贝提卡假单胞菌、诺丽鞘氨醇单胞菌、皮尔瑞俄类芽孢杆菌、巨黑木霉、黑褐木霉、非洲哈茨木霉和云南木霉具备开发成微生物防治剂的潜力。
- Abstract:
- Taking stems and leaves of diseased roses from Nanyang as test materials,pathogenic fungi were isolated through the tissue separation method and identified using molecular biology techniques.Antagonistic bacteria were screened via plate confrontation assays,and their pathogenicity was assessed in vitro to identify the causative fungal species and screen antagonistic bacteria,in order to provide references for microbial control strategies targeting early-stage fungal diseases in roses.The results showed that,based on morphological characteristics and ITS sequence analysis,the pathogenic fungi were Colletotrichum aenigma(B8),Botryotinia ranunculi(B14),Nigrospora musae(B39),and Fusarium longifundum(B44).Pathogenicity tests confirmed that all four fungal strains exhibited pathogenic effects on rose plants.Pseudomonas baetica, Sphingomonas morindae,and Paenibacillus peoriae exhibited an antibacterial rate exceeding 90% against the four pathogens.Fungal antagonism experiments revealed varying inhibitory effects of Trichoderma melanomagnum,Trichoderma atrobrunneum,Trichoderma afroharzianum,and Trichoderma yunnanense on the four pathogens.These strains displayed significant inhibition against B8,B39,and B44;however,their inhibitory effects on B14 were not satisfactory.To sum up,the potential of Pseudomonas baetica,Sphingomonas morindae,Paenibacillus peoriae,Trichoderma melanomagnum,Trichoderma atrobrunneum,Trichoderma afroharzianum and Trichoderma yunnanense as promising candidates for the development of microbial control agents.
