枸杞扦插生根及相关生理生化分析

- 以6份枸杞无性系为试材,采用相同IBA浓度处理,分析插穗的生根过程以及生根过程中营养物质、酶活性等含量的变化,以期探究枸杞生根的作用机理,促进枸杞在生产实践中的推广应用。结果表明:HT2017-01、HT2017-02、HT2017-05和HT2017-06的生根类型为愈伤组织生根型,HT2017-03和HT2017-04为皮部生根型。6份枸杞无性系中,生根率、平均根数和生根指数最大的均为HT2017-02,显著优于其它无性系,平均根长最长的为HT2017-05。插穗内营养物质和氧化酶活性变化各不相同。6份枸杞无性系在扦插过程中随着愈伤组织的形成,可溶性糖含量(除HT2017-03)、可溶性蛋白质含量和多酚氧化酶(PPO)活性均升高;在不定根伸长阶段,HT2017-01、HT2017-03和HT2017-04的可溶性糖含量上升,HT2017-02、HT2017-05和HT2017-06的可溶性糖含量下降,可溶性蛋白质含量和PPO活性均升高。过氧化物酶(POD)除HT2017-04外,其它均呈现先上升后降低、再上升降低的趋势,呈现双峰曲线的形式。

- Six Lycium barbarum clones were used as experimental materials and treated with the same IBA concentration to analyze the rooting process of cuttings and the changes in the contents of nutrients and enzyme activities during the rooting process,in order to explore the mechanism of rooting and promote the promotion and application of Lycium barbarum in production practice.The results showed that the rooting types of HT2017-01,HT2017-02,HT2017-05 and HT2017-06 were callus rooting type,and HT2017-03 and HT2017-04 were skin rooting type.Among the six Lycium barbarum clones,HT2017-02 had the highest rooting rate,average root number and rooting index,which was significantly better than other clones,and HT2017-05 had the longest average root length.The changes of nutrient and oxidase activities in cuttings were different.The soluble sugar content (except HT2017-03),soluble protein content and PPO activity of six Lycium barbarum clones increased with the formation of callus during cuttage.In adventitious root elongation stage,the soluble sugar content of HT2017-01,HT2017-03 and HT2017-04 increased,while the soluble sugar content of HT2017-02,HT2017-05 and HT2017-06 decreased,while the soluble protein content and PPO activity increased.Except for HT2017-04,all POD showed a trend of rising first,then decreasing,and then rising and decreasing,presenting a form of bimodal curve.
