欧李叶片光合色素提取方法的比较分析

- 以‘宁欧’、蒙原欧李和‘农大7号’3个抗旱性不同的欧李叶片为试材,通过比较分析4种提取方式6种提取溶剂对叶绿素a、叶绿素b和类胡萝卜素3种光合色素的提取效果,筛选出适合欧李叶片光合色素提取的最佳方案,以期为欧李光合生理、品种选育及抗性生理等方面的研究提供理论参考依据。结果表明:4种提取方法中,研磨法提取效率最高,优选提取试剂C(丙酮∶乙醇=1∶1);干样浸提法效率低,但受品种影响小,优选提取试剂B(95%乙醇);鲜样浸提法提取效率较高,但受品种抗性影响较大。液氮法在保存和研磨过程中色素发生降解,含量较低。
- Abstract:
- ‘Ningou’,Mengyuan and ‘Nongda No.7’ were used as experimental materials,and the extraction effects of six extraction solvents under four extraction methods on photosynthetic pigments including chlorophyll a,chlorophyll b and carotenoid were compared and analyzed,the optimal extraction scheme for photosynthetic pigment in leaves of three varieties of Chinese dwarf cherry was screened out,in order to provide a reference for the study of photosynthetic physiology,variety breeding and resistance physiology of Cerasus humilis.The results showed that among the four extraction methods,grinding method had the highest extraction efficiency,and the optimum extraction reagent C (acetone∶ethanol 1∶1) was selected.The dry sample extraction method had low efficiency,but it was less affected by the variety.The optimum extraction reagent B (95% ethanol) was selected.The extraction efficiency of fresh sample extraction method was higher,but it was influenced by the resistance of varieties.The pigment was degraded under liquid nitrogen method during storage and grinding,and the content was low.
