- 以低温湿沙贮藏71 d的野生毛冬青(Ilex pubescens)种子为试验材料,设置3个温度组(20、25、30 ℃),6个浸种溶液组(5、10、20 g?L-1赤霉素(GA3)以及5 g?L-1硝酸钾(KNO3)、5 g?L-1 KNO3加10 g?L-1 GA3、对照组)进行萌发试验,以发芽率、发芽势、发芽指数和活力指数等为评价指标,考察各组毛冬青种子萌发和幼苗生长情况,以期筛选出适宜毛冬青种子萌发的最佳温度条件和浸种溶液浓度。结果表明:3个温度条件下毛冬青种子发芽率无显著差异,20 ℃时种子的发芽势和发芽指数均显著低于25 ℃组;30 ℃组种子的活力指数显著低于25 ℃组。用10 g?L-1GA3溶液浸种能显著促进种子萌发,使发芽势和发芽指数等评价指标显著高于对照组(蒸馏水浸种);5 g?L-1KNO3溶液浸种24 h对毛冬青种子萌发及其幼苗的生长无明显影响。因此,25 ℃是最适宜毛冬青种子萌发的温度,10 g?L-1GA3是最适宜的浸种溶液浓度。
- Abstract:
- To seek a proper temperature for Ilex pubescens seeds germination and find an optimal concentration of gibberellin acid(GA3) or potassium nitrate(KNO3) for seeds pretreatment.After stored with wet sand at 4 ℃ for 71 days,seeds were set up in 8 groups and sowed in petri dishes.There were 3 groups for temperature investigation,20 ℃,25 ℃ and 30 ℃.And 5,10,20 g?L-1 GA3,5 g?L-1 KNO3,mixed solution of 5 g?L-1 KNO3 and 10 g?L-1 GA3,and distilled water were used to pre-soak the seeds respectively.Parameters of germination percentage,germination energy,germination index,fresh weight of seedlings,vigor index,lengths of radical and hypocotyl were used to investigate the effects.There was no significant difference of the germination percentages between the groups at three temperatures;germination energy and germination index of seeds at 20 ℃ were significantly lower than those in 25 ℃.Vigor index of seeds at 30 ℃ was significantly lower than those in 25 ℃.After soaking seed with 10 g?L-1GA3,the germination energy and germination index were significantly higher than the control group.Conclusion the optimal germination temperature was 25 ℃,and 10 g?L-1 GA3 was suitable for pretreatment of Ilex pubescens seeds before germination.
