滇牡丹类黄酮5-O-葡萄糖基转移酶基因的鉴定及表达分析

- 以野生黄花滇牡丹(Paeonia delavayi)为试材,采用RT-PCR技术,克隆得到一个具完整开放阅读框的类黄酮5-O-葡萄糖基转移酶([STBX]Pd5GT[STBZ])基因,并对其进行生物信息学分析和转录模式分析,以期为研究滇牡丹花色素形成机理,滇牡丹糖基转移酶功能鉴定及花卉品质改良提供理论基础。结果表明:该基因全长1 410 bp,编码469个氨基酸(GenBank登录号:ARA67364);生物信息学分析发现Pd5GT蛋白C末端含有PSPG盒子,其氨基酸序列为WCSQVEVLSHPSLGCFVTHCGWNSTTESLVCGVPVVAFPQWTDQGTNAKL,与已知功能的5GT蛋白序列聚为一类;该基因在茎、叶中微弱表达,在花中大量表达,在不同颜色的花瓣中均可大量表达。综上所述[STBX]Pd5GT[STBZ]基因可能编码5-O-糖基转移酶,具有一定的组织特异性,花色特异性不明显。

- Paeonia delavayi was used as experimental materials,we cloned a full length of flavonoid 5-O-glucosyltransferase([STBX]Pd5GT[STBZ]) gene from P.delavayi firstly with the method of RT-PCR,which has a whole opening reading frame,and used the bioinformatics and transcriptional pattern to analyze [STBX]Pd5GT[STBZ] gene.The results showed that the C terminal of Pd5GT protein had PSPG box,and its amino acids sequence was WCSQVEVLSHPSLGCFVTHCGWNSTTESLVCGVPVVAF PQWTDQGT NAKL,and clustered together with 5GT known their function;[STBX]Pd5GT[STBZ] gene was strongly expressed in flower,slightly in stem and leaves,and strongly expressed in different color petals.In conclusion,[STBX]Pd5GT[STBZ] gene could encode 5-O-glucosyltransferase,had some tissue-speciality but flower-color-speciality wasn′t obviously.The present study would provide theoretical basis for the study of mechanism of flower pigment biosynthesis in P.delavayi,for the identification of the function of glucosyltransferase in P.delavayi,and for improving flower quality by genetic engineering.
