- 以当年成熟的‘白兰地’海棠(Malus ‘Brandywine’)种子为试材,通过种子萌发试验、种子形态观察、种皮透水性测定、种皮和种胚内源抑制物的生物测定、种皮内源抑制物分离、GC-MS技术(气相色谱-质谱联用技术)分析,初步研究了‘白兰地’海棠种子的休眠特性,以期为促进海棠新品种培育提供参考依据。结果表明:‘白兰地’海棠种子不存在形态后熟,种胚具有浅度生理休眠,种子萌发时不存在种皮透水障碍。‘白兰地’海棠的种皮甲醇浸提液对黑麦草(Lolium perenne)和白菜(Brassica pekinensis)种子萌发与幼苗生长的抑制作用强于种胚浸提液,种皮所含内源抑制物是‘白兰地’海棠种子长时间休眠的主要因素。‘白兰地’海棠的种皮各分离相对白菜种子发芽的抑制作用由弱到强依次为水相<石油醚相<乙酸乙酯相<甲醇相<乙醚相。对‘白兰地’海棠种皮各分离相进行了GC-MS分析,发现‘白兰地’海棠的种皮中主要存在亚油酸等有机酸类、油酸酰胺等酰胺类、2,6-二叔丁基对甲酚等酚类等有机化合物。其中,种皮所含的有机酸类、酚类等内源抑制物质可能是‘白兰地’种子休眠的主要因素。
- Abstract:
- The seeds of Malus ‘Brandywine’ which matured in the current year were taken as the test materials.Through germination tests,seed morphological observations,determinations of the water permeability of seed coats,bioassays of germination inhibitor in seed coats and embryos,separations of germination inhibitor in seed coats,and combined with Gas Chromatography-Mass Spectrometry,the dormancy characteristics of M.‘Brandywine’seeds were studied,in order to provide reference for breeding of new varieties of Malus.The results showed that the seeds of M. ‘Brandywine’ didn′t have morphological after-ripening,the embryos had a slight physiological dormancy,and there was no water permeability barrier of the seed coat during seed germination.The methanol extract of the seed coat of Malus ‘Brandywine’ had a stronger inhibitory effect on the seed germination and seedling growth of Lolium perenne and Brassica pekinensis than that of the seed embryo.The germination inhibitor contained in the seed coat was the main factor for the long-term dormancy of the seeds of M. ‘Brandywine’.The inhibitory effects of phase extracts of the seed coat of M. ‘Brandywine’ on the germination of Brassica pekinensis seeds,arranged from weak to strong,was as follows,water phase
