- 以采自黑龙江省伊春市的野生硫磺菌2023V48菌株为试材,采用组织分离法获得其母种,通过形态学特征和分子生物学鉴定法对其进行鉴定,并采用单因素和正交实验,研究了碳源、氮源、无机盐等对菌丝体生长的影响,同时开展驯化栽培试验,以期为硫磺菌的栽培应用提供参考依据。结果表明:在单因素试验中硫磺菌菌丝生长的最适碳源为玉米粉,最适氮源为麸皮,最适无机盐硫酸亚铁,最适pH为5.0,最适培养温度为25~28 ℃。根据单因素试验结果,设计了正交实验,得出适合菌丝生长最佳组合为A2B2C1D3,即玉米粉为20 g·L-1、麸皮为2.0 g·L-1、FeSO4为0.5 g·L-1、NaCl为1.5 g·L-1。各因素对菌丝生长的影响程度顺序为NaCl>FeSO4>麸皮>玉米粉。
- Abstract:
- Taking strain isolated from wild Laetiporus sulphureus 2023V48 fruitbody collected from Yichun city,Heilongjiang Province as the experimental material,it was identified by morphological characteristics and molecular biological identification method.The effects of carbon source,nitrogen source and inorganic salt on the growth of mycelia were studied by single factor and orthogonal experiments.And the domestication and cultivation experiments were conducted, in order to provide reference for the cultivation and application of Laetiporus sulphureus.The results showed that the optimum carbon source was corn flour,the optimum nitrogen source was wheat bran,the optimum inorganic salt was ferrous sulfate,the optimum pH was 5.0,and the optimum culture temperature was 25-28 ℃.According to the results of single factor experiment,the orthogonal experiment was designed,and the best combination for mycelial growth was A2B2C1D3,that was,corn flour was 20 g·L-1,wheat bran was 2.0 g·L-1,FeSO4 was 0.5 g·L-1,NaCl was 1.5 g·L-1.The influence degree of each factor on mycelial growth was in the order of NaCl>FeSO4>wheat bran>corn flour.
