- 以野生大球盖菇酒红色菌株及其白色和黄色突变株为试材,采用单因素试验和正交实验,筛选最适母种培养基,以期为大球盖菇优良菌株的利用提供参考依据。结果表明:3个菌株间存在明显拮抗反应,单因素试验表明酒红色菌株和白色突变株最适碳源为蔗糖,最适氮源为蛋白胨,最佳无机盐为氯化钙和硫酸钾。黄色突变株最适碳源为可溶性淀粉和蔗糖,最适氮源为麸皮和酵母膏,最适无机盐为硫酸钾,最适维生素为VB2。黄色突变株菌丝生长速度最快,长势旺盛、洁白浓密。正交实验确定黄色突变株最优培养基配方为马铃薯200 g·L-1、干稻草100 g·L-1、蔗糖15 g·L-1、可溶性淀粉5 g·L-1、酵母膏1 g·L-1、麸皮1.5 g·L-1、硫酸钾1 g·L-1、维生素B2 10 mg·L-1,菌丝生长速度为2.27 mm·d-1。综上,3个菌株在不同培养基上菌丝生长情况均存在一定差异,稻草浸出液能促进菌丝生长,黄色突变株培养时间较PDA培养基缩短6 d。
- Abstract:
- The wild burgundy strain of Stropharia rugosoannulata and its white and yellow mutant strains were used as the test materials to screen the most suitable mother culture medium using one-way and orthogonal tests,in order to provide reference for the utilization of excellent strains of Stropharia rugosoannulata.The results showed that there was a significant antagonistic reaction among the three strains,and the one-way test indicated that the optimal carbon source for the burgundy strain and the white mutant was sucrose,the optimal nitrogen source was peptone,and the optimal inorganic salts were calcium chloride and potassium sulphate.The optimal carbon sources for the yellow mutant strain were soluble starch and sucrose,the optimal nitrogen sources were bran and yeast paste,the optimal inorganic salt was potassium sulphate,and the optimal vitamin was vitamin B.The yellow mutant strain had the fastest growing mycelium,which was vigorous,white and dense.Orthogonal tests determined that the optimal medium formulation for the yellow mutant strain was 200 g·L-1 potato,100 g·L-1 dried rice straw,15 g·L-1 sucrose,5 g·L-1 soluble starch,1 g·L-1 yeast paste,1.5 g·L-1 bran,1 g·L-1 potassium sulphate,and 10 mg·L-1 vitamin B,and the mycelial growth rate was 2.27 mm·d-1.There were some differences in mycelial growth of all three strains on different media,and the rice straw extract promoted mycelial growth,and the incubation time of the yellow mutant strain was shortened by 6 days compared with that of the PDA medium.
