- 以13个省市的106份党参种质资源为试材,采用植物形态和DNA分子鉴定相结合的方法,研究了106份党参种质资源间的亲缘关系,以期为植物形态特征和DNA分子技术鉴定党参种质资源提供参考依据。结果表明:党参、川党参、素花党参与管花党参、羊乳在植物形态上有较大的差异,而党参、川党参和素花党参之间差异较小,但均可以通过植物形态差异进行鉴别;利用ITS2序列和psbAtrnH序列可准确鉴别党参、川党参、素花党参与管花党参、羊乳,但对变种与原变种间的鉴定仍存在一定局限性。
- Abstract:
- Taking 106 Codonopsis resources from 13 provinces and cities as the test materials,the genetic relationship of 106 Codonopsis germplasm resources was studied by the combination of plant morphological characteristics and DNA molecular identification,in order to provide reference for identifying Codonopsis germplasm resources by DNA barcoding combined with plant morphological characteristics.The results showed that there were significant morphological differences among C.pilosula (Franch.) Nannf., C.tangshen Oliv.,C.pilosula Nannf.var.modesta (Nannf.) L.T.Shen as well as C.tubulosa,C.lanceolata.Differences between C.pilosula (Franch.) Nannf., C.tangshen Oliv.and C.pilosula Nannf.var.modesta (Nannf.) L.T.Shen were smaller,but all could be identified by plant morphological differences.ITS2 and psbAtrnH sequences accurately identified C.pilosula (Franch.) Nannf., C.tangshen Oliv.,C.pilosula Nannf.var.modesta (Nannf.) L.T.Shen as well as C.tubulosa,C.lanceolata.However,limitations were encountered in identifying variant species among the original variants.
