- 以表现黄色环纹症状的8份茉莉叶片为试材,采用RT-PCR方法,研究了待测茉莉的病毒种类及茉莉T病毒(jasmine virus T,JaVT)外壳蛋白(coat protein,CP)基因序列遗传多样性,以期为茉莉病毒病的防治提供参考依据。结果表明:4份样品中检测到JaVT,并扩增出4条JaVT的外壳蛋白(CP)基因序列。4个JaVT分离物CP基因间核苷酸序列的一致性为92.20%~95.20%,与NCBI已发表的4个JaVT CP基因核苷酸序列一致性为91.40%~94.00%。基于8个JaVT CP基因全长序列进行系统发育分析,结果表明该研究中的4个JaVT分离物与来自中国的JaVT-MZ962345聚集在一起。8条JaVT全长CP基因序列中检测到3个重组事件。这是甘肃省首次报道茉莉T病毒。
- Abstract:
- Eight jasmine leaves showing yellow ring were used as experimental material.And the pathogen causing jasmine virus disease and the nucleic acid sequence,genetic diversity of jasmine virus T (JaVT) were analyzed by RT-PCR,in order to provide reference for the prevention and treatment of jasmine virus disease.The results showed that JaVT was isolated from 4 samples,and 4 CP gene sequences of JaVT were amplified.The nucleotide sequence identity among the CP genes of the four JaVT isolates was 92.20%-95.20%,and the nucleotide sequence identity between the CP genes of the four JaVT isolates was 91.40%-94.00% with the other 4 JaVT CP genes published in NCBI.Phylogenetic analysis based on the full-length sequences of 8 JaVT CP genes.The results showed that the four JaVT isolates in this study were clustered with JaVT-MZ962345 from China.Three recombination events were detected in 8 JaVT full-length CP gene sequences.
