- 以河西地区羊肚菌白霉病的病原为试材,采用组织分离法对羊肚菌白霉病病原进行分离纯化,通过形态学和分子生物学方法对病原进行鉴定,研究了不同温度、pH、碳源、氮源和培养基对病原菌生长的影响,以期为制定羊肚菌白霉病有效防控措施提供参考依据。结果表明:结合形态学和ITS(internally transcribed spacer)序列分析,羊肚菌白霉病的病原为长孢卵单隔孢霉(Diploospora longispora);生物学特性研究表明,病原菌适宜生长温度范围为20~25 ℃,在30 ℃以上停止生长,在酸性环境中长势较好,最适生长pH为6.0~6.5;最适生长碳、氮源和培养基分别为蔗糖、蛋白胨和PCA培养基。
- Abstract:
- Taking the pathogen of Morchella white mold disease in Hexi Area as the experimental materials,the pathogen of Morchella white mold was isolated and purified by tissue separation method.The pathogen was identified by morphological and molecular biological methods,and the effects of different temperature,pH,carbon source,nitrogen source and medium on the growth of the pathogen were clarified,in order to provide reference for the development of effective prevention and control measures of morel of Morchella white mold.The results showed that the pathogen was Diploospora longispora based on morphological and ITS sequence analysis.The biological characteristics showed that the suitable growth temperature range of the pathogen was 20-25 ℃,and the pathogen stopped growing above 30 ℃.The pathogen grew well in acidic environment,and the optimum growth pH was 6.0-6.5.The optimum carbon source,nitrogen source and medium for growth were sucrose,peptone and PCA medium,respectively.
