- 以工厂化栽培海鲜菇菌包为试材,采用韦恩分析、偏最小二乘判别分析、聚类分析和通路富集分析等方法,利用非靶向代谢组学研究不同培养时期海鲜菇菌包的代谢产物动态变化特征,分析不同培养时期海鲜菇菌包的代谢产物,以期为阐明海鲜菇代谢调控机制和优化海鲜菇菌包提供参考依据。结果表明:不同培养时期海鲜菇菌包共鉴定出3 205种代谢产物,其中不同培养时期海鲜菇菌包中均共有代谢产物1 844种(87.4%),主要包含羧酸及其衍生物、脂肪酰基、有机氧化合物、异戊醇脂质等物质。代谢途径富集分析表明,氨基酸类代谢、碳水化合物代谢、辅因子和维生素的代谢、核苷酸代谢、脂质代谢等途径占据主导地位。通过与对照(未接种菌包)相比,筛选出121种差异代谢物,隶属于15条代谢途径,其中赖氨酸生物合成,辅因子的生物合成,核黄素代谢,缬氨酸、亮氨酸和异亮氨酸生物合成,泛酸和辅酶A生物合成,硫胺素代谢等6条代谢途径表现出显著性差异(P<0.05),它们主要属于氨基酸类和B族维生素类代谢产物,而且这些代谢途径对海鲜菇子实体、营养物质和鲜味形成具有重要影响。不同培养时期海鲜菇菌包代谢产物表现出明显差异,表现出培养早期(Hm2)和出菇期(Hm5)特有代谢物变化较大,培养中期(Hm4)代谢物变化则相对较少。该研究从代谢组学角度揭示了海鲜菇菌包培养过程中代谢产物动态变化特征及其生物合成的潜在机制。
- Abstract:
- The dynamic changes of metabolites in H.marmoreus during different culture periods were studied by untargeted metabolomics industrially using cultivated H.marmoreus as experimental materials.The metabolites of H.marmoreus in different culture periods were analyzed throughout Venn analysis,partial least squares discriminant analysis,cluster analysis and pathway enrichment analysis.The results showed that a total of 3 205 metabolites were identified in H.marmoreus during different culture periods.Among them,there were 1 844 metabolites (87.4%) in H.marmoreus during different culture periods.Mainly included carboxylic acids and their derivatives,fatty acyls,organic oxygen compounds,prenol lipids.Herein Metabolic pathway enrichment analysis showed amino acid metabolism,carbohydrate metabolism,cofactor and vitamin metabolism,nucleotide metabolism,lipid metabolism and other pathways dominated.Compared with the control group(without inoculation),121 differential metabolites were screened,belonging to 15 metabolic pathways.Among them,there were a series of chemicals showed striking features,for instance,lysine biosynthesis,cofactor biosynthesis,riboflavin metabolism,valine,leucine and isoleucine biosynthesis,pantothenic acid and coenzyme A biosynthesis,thiamine metabolism and other 6 metabolic pathways showed significant differences (P<0.05),mainly belong to amino acids and B vitamin metabolites,and apart from that,these metabolic pathways display have important effects on the formation of fruiting bodies,nutrients and umami of H.marmoreus.The metabolites of H.marmoreus showed significant differences in different cultivation periods,which tells the specific metabolites in the early stage of culture (Hm2) and fruiting stage (Hm5) changed greatly,while the metabolites in the middle stage of culture (Hm4) changed little compared with those two.This study revealed the dynamic changes of metabolites and the potential mechanism of biosynthesis during the cultivation of H.marmoreus from the perspective of metabolomics,and also provided a scientific basis for elucidating the metabolic regulation mechanism of H.marmoreus and optimizing H.marmoreus.
