- 以收集的28份柴胡种子为试材,采用种子外观鉴定和DNA分子鉴定相结合的方法,研究了28份柴胡种子间的亲缘关系,以期为DNA条形码和种子的形态特征结合来鉴定柴胡种子提供参考依据。结果表明:利用ITS2分析北柴胡种子的序列聚为一支,黑柴胡聚为一支,抱茎柴胡与窄竹叶柴胡聚为一支,有很好的单系性,与柴胡种子形态特征的分析结果一致。
- Abstract:
- Taking 28 Bupleurum chinense seeds as test materials,the genetic relationship of 28 Bupleurum chinense seeds was studied by the combination of seed appearance identification and DNA molecular identification,in order to provide reference for identifying Bupleurum chinense seeds by DNA barcoding combined with seed morphological characteristics.The results showed that with ITS2 analysis the seed sequences of Bupleurum chinense,Bupleurum smithii were clustered into one branch separately,B.longicaule var.amplexicaule and B.marginatum var.stenophyllum were clustered into one branch,which had good monophyly.The morphological characteristics of Bupleurum chinense seeds were consistent with ITS2.The research provided references for identifing Bupleurum chinense seeds by DNA barcoding combined with seed morphological characteristics.
