- 以一株巴布亚新几内亚野生香菇为试材,采用ITS、LSU和SSU序列分析对其进行分子鉴定,并采用单因素试验和正交实验探究其菌丝生物学特性。以期明确其分类学地位以及为砖红小香菇Lentinula lateritia的人工栽培和后续的深入研究提供参考依据。结果表明:该野生香菇的ITS序列长度为725 bp,LSU序列长度为934 bp,SSU序列长度为1 100 bp,均与砖红小香菇相似度最高,鉴定该菌株为砖红小香菇Lentinula lateritia;生物学特性研究结果表明,该野生香菇菌丝生长的最适碳源为果糖,最适氮源为牛肉膏,最适pH为4.0,最适生长温度25 ℃。
- Abstract:
- A wild Lentinula edodes from Papua New Guinea was used as test material,we used sequence analysis of ITS,LSU and SSU for molecular identification,and used single factor tests and orthogonal tests to explore its mycelial biological characteristics.In order to clarify the taxonomic status of the wild Lentinula edodes and provide a reference for the artificial cultivation and further research of Lentinula lateritia.The results showed that the length of the ITS sequence was 725 bp,the length of LSU sequence was 934 bp,and the length of SSU sequence was 1 100 bp,which had the highest similarity with Lentinula lateritia.The strain was identified as Lentinula lateritia.The results of biological characteristics showed that the optimal carbon source for the growth of wild Lentinula edodes mycelial was fruticose,the optimal nitrogen source was beef extract,the optimal pH was 4.0,and the optimal growth temperature was 25 ℃.
