- 以连翘花粉为试材,采用液体培养法,研究了培养基中蔗糖、氯化钙、硼酸、PEG及不同培养时间对连翘花粉萌发和花粉管生长的影响,通过醋酸洋红和TTC染色法,找出可快速测定连翘花粉活力的方法,以期为连翘杂交育种提供参考依据。结果表明:蔗糖、硼酸、氯化钙和PEG显著影响连翘花粉的萌发和生长,离体萌发最佳培养基组合为15%蔗糖+0.015%硼酸+0.020%氯化钙+10%PEG,最适培养时间为3 h,萌发率可达87.20%,花粉管长度达181.20 μm;花粉活力测定中,醋酸洋红染色法适用于花粉活力快速测定,和离体萌发结果基本一致。
- Abstract:
- Pollen of Forsythia suspensa was used as the experimental material,the effects of sucrose,CaCl2,H3BO3,PEG and different culture times on pollen germination and pollen tube growth of Forsythia suspensa were studied by using pollen liquid culture.By comparing in vitro pollen germination culture method,TTC staining method and acetic acid magenta staining method,the suitable method for rapid determination of pollen vitality of forsythia was obtained,in order to provide a reference for cross breeding of Forsythia suspensa.The results showed that,the concentrations of,sucrose CaCl2,H3BO3 and PEG in the medium had significant influence on pollen germination and pollen tube growth,the optimal medium combination for in vitro germination of forsythia pollen was 15% sucrose+0.015% H3BO3+0.020% CaCl2+10% PEG,the optimal culture time was 3 hours,the higher pollen germination rate and longest pollen tube reached 87.20% and 181.20 μm respectively.Acetic acid magenta staining method can be used for the rapid determination of pollen vitality of forsythia,resembling the result of in vitro germination.
