- 以香菇L808为试材,采用单因素试验和正交实验相结合的方法,优化香菇母种培养基,以期为香菇产业发展提供更好的技术依据。结果表明:细木屑90 g?L-1、葡萄糖20 g?L-1、琼脂粉18 g?L-1、去皮马铃薯200 g?L-1为香菇母种最佳培养基。利用该配方培养的香菇母种,用于制作原种时菌丝分解纤维素的能力增强,菌丝洁白、粗壮、浓密,菌丝生长速度较未优化的母种培养基提高30.2%,显著提高香菇菌种质量。而母种配方为细木屑120 g?L-1、葡萄糖10 g?L-1、琼脂粉20 g?L-1、去皮马铃薯190 g?L-1比较适宜进行香菇母种斜面保藏,菌种菌丝生长速度较慢,菌丝洁白粗壮,有效保持菌丝活力,防止菌种退化。
- Abstract:
- Using Lentinu sedodes strain L808 as test material,a combination of single factor test and orthogonal test was used to optimize the culture medium of Lentinu sedodes stock culture.The results showed that 90 g?L-1 of sawdust,20 g?L-1 of glucose,18 g?L-1 of agar powder,and 200 g?L-1 of peeled potatoes were the best culture medium for the parent species of Lentinu sedodes stock culture.The stock culture of Lentinu sedodes cultivated with the formula has enhanced mycelial ability to decompose cellulose when used to make the original seed.The mycelium was white,thick and dense,and the growth rate of mycelium was increased by 30.2% compared with the unoptimized parent medium.The quality of culture Lentinu sedodes was significantly improved.The formula of the stock culture was 120 g?L-1 of sawdust,10 g?L-1 of glucose,20 g?L-1 of agar powder,and 190 g?L-1 of peeled potatoes.It was more suitable for the oblique storage of the Lentinu sedodes stock culture.The growth rate of the mycelium was relatively slow,and the mycelium was white and thick,which effectively maintains the viability of the mycelium and prevents the degradation of the variety.
