金叶杨组织培养再生体系的建立

- 以金叶杨(Populus nigra cv.‘Jinye’)带腋芽的茎段为试材,采用了组织培养的方法,研究了金叶杨的腋芽诱导、继代增殖、生根培养,以期为保持其优良性状、实现工厂化育苗提供参考。结果表明:1年生茎段最佳消毒方式是使用0.1%HgCl2溶液处理10 min;最佳诱导培养基为MS培养基;腋芽诱导最佳激素配比为6-BA 0.5 mg·L-1+NAA 0.3 mg·L-1,腋芽诱导率为92.6%;增殖效果最佳的激素配比为6-BA 1.0 mg·L-1+NAA 0.4 mg·L-1+GA3 0.2 mg·L-1,增殖系数为4.4;组培苗生根培养效果最好的培养基为1/2MS+IBA 0.5 mg·L-1,生根率为91.2%。
- Abstract:
- The stem segment with axillary buds of Populus nigra cv.‘Jinye’ were used as explant,the method of tissue culture was adopted,through the selection of the most suitable culture medium,the axillary bud induction,subculture proliferation and rooting were studied,in order to provide the basic research theory for maintaining its excellent characters and realizing the factory seedling.The results showed that the best disinfection method for annual stems was 0.1%HgCl2 for 10 minutes.For axillary bud induction using stem,the highest induction frequency of axillary bud was 92.6%,using MS containing 6-BA 0.5 mg·L-1 and NAA 0.3 mg·L-1.The best hormone ratio for proliferation was 6-BA 1.0 mg·L-1 and NAA 0.4 mg·L-1,additional GA3 0.2 mg·L-1.On this medium,the multiplication coefficient could reach to 4.4,we inducted seedling rooting on 1/2MS and 0.5 mg·L-1 IBA,the rooting rate was 91.2%.
