- 柚喀木虱Cacopsylla citrisuga Yang&Li是柑桔黄龙病的传播媒介,在云南省宏德州主要为害柠檬、柚子等果树类的嫩梢及嫩叶,其成虫及若虫体内均检测到黄龙病亚洲种病原菌‘Candidates Liberibacter asiaticus’。该研究基于16S rDNA基因的RFLP指纹图谱法,建立了野外柠檬和柚子上柚喀木虱成虫体内微生物的16S rDNA文库,分析了柚喀木虱体内共生菌多样性。结果表明:柚喀木虱体内存在共9种体内共生菌,木虱/喀木虱次生共生菌(secondary endosymbiont of Psylla/secondary endosymbiont of Cacopsylla)、不可培养菌(uncultured bacterium)、Arsenophonus endosymbiont、Pantoea sp.、uncultured Erwinia sp.、Pseudomonas sp.、Rahnella sp.、Enterobacter sp.、Wolbachieae bacterium。根据指纹图谱结果分析,每个文库所分布操作单元(OUT)结果均不同,表明不同寄主植物及性别的柚喀木虱成虫体内共生菌的种类及数量存在差异。
- Abstract:
- Cacopsylla citrisuga Yang&Li was an insect vector of huanglongbing.This pest damaged the young leaves and flushes of lemon and pummel trees in Hongde Yunnan Province.A recent study indicated that both nymphs and adults of C.citrisuga carried ‘Candidates Liberibacter asiaticus’,the bacterium associated with huanglongbing.In this study,the endosymbiont 16S rDNA library of C.citrisuga collected from lemon and pomelo trees was built based on the RFLP of 16S rDNA gene,and the diversity of the endosymbionts was analyzed.The results showed that there were nine kinds of endosymbiont in this pest species,including secondary endosymbiont of Psylla/secondary endosymbiont of Cacopsylla,uncultured bacterium,Arsenophonus endosymbiont,Pantoea sp.,uncultured Erwinia sp.,Pseudomonas sp.,Rahnella sp.,Enterobacter sp.and Wolbachieae bacterium.According to the RFLP,each of the OUT was just different from others,indicated that species and quantity of the endosymbionts were different in C.citrisuga of different postplant and gender.
