- 以洋紫荆种子为试验材料,采用60Coγ辐照、种子预处理和发芽试验等方法,研究了不同预处理洋紫荆种子对60Coγ辐照诱变的敏感程度,以期为洋紫荆的辐照诱变育种提供理论和技术基础。结果表明:洋紫荆干燥、冷藏和浸泡6 h种子,在辐照剂量超过或等于120 Gy时,相比低辐照强度下,萌芽率由89.33%~92.33%下降到72.00%~79.67%、萌芽指数由158.00~174.80下降到82.50~106.30、出苗率由74.33%~75.33%下降到50.00%~55.67%、出苗指数由84.73~88.40下降到24.63~37.50、成苗率由70.00%~72.33%下降到42.00%~46.00%、苗高由25.80~29.03 cm下降到18.00~23.13 cm、叶片开角变化率由2.67%~5.00%上升到9.67%~17.00%,种子萌发和幼苗生长的大多数指标出现显著变化;而浸泡12 h种子,在辐照剂量超过或等于60 Gy时,种子和幼苗的大多数指标就显示出类似显著改变。因此,当洋紫荆种子含水量升高或解除休眠时,其对60Coγ辐照的敏感程度会提高,反之则敏感程度会降低。这将为不同保存或预处理条件下洋紫荆种子的辐照诱变提供一定的基础。
- Abstract:
- The seeds of Bauhinia were used as materials, 60Coγ irradiation,seed pretreatment and germination experiment methods were used,the sensitivity of Bauhinia variegata seeds to the radiation-induced mutation of 60Coγ was studied,in order to provide theoretical and technical basis for the radiation mutation breeding of Bauhinia variegata.The results showed that,under a greater than or equal to 120 Gy irradiation dose,the germination rate decreased from 89.33%-92.33% to 72.00%-79.67%,the germination index from 158.00-174.80 to 82.50-106.30,the seedling rate from 74.33%-75.33% to 50.00%-55.67%,the seedling rate from 84.73-88.40 to 24.63-37.50,the seedling rate from 70.00%-72.33% to 42.00%-46.00%,the seedling height from 25.80-29.03 cm to 18.00-23.13 cm and the change rate of blade opening angle increased from 2.67%-5.00% to 9.67%-17.00%,suggesting the beginning of significant changes in most indicators of seeds and seedlings.However,the majority of the indicators of the seeds and seedlings had begun to show similar significant changes after being soaked for 12 hours seeds under over or equal to 60 Gy irradiation dose.Therefore,when the water content of Bauhinia variegata seeds was increased or the dormancy is broken,their sensitivity to the radiation of the 60Coγ would become higher;otherwise,the sensitivity would become lower.The conclusion would lay a solid foundation for the irradiation induced mutation of the Bauhinia variegata seeds under different preservation or pretreatment conditions.
