额敏贝母种子休眠原因的初步研究

- 以新疆额敏贝母种子为试材,采用扫描电镜观察种子形态、TTC法测定种子活力、烘箱称重法测量种子含水量、浸种称重法测定种子透水性、体视显微镜观察胚变化及萌发情况、生物鉴定法检测发芽抑制物类型,研究了额敏贝母种子休眠原因及其特殊性,确定种子休眠类型,以期为该种的引种驯化提供参考。结果表明:额敏贝母种皮薄如蝉翼,种翅密布小气孔,吸水速率比川贝母更快,2 h内种子吸水率就可以达到100%以上,不存在吸水障碍。胚率的变化及种子发芽情况得出胚发育不完全存在形态后熟和生理后熟。因此得出额敏贝母种皮透水性,透气性不是引起种子休眠的直接原因。额敏贝母不属于具有成熟胚的贝母属植物,因种胚发育不完全导致形态休眠及萌发抑制物导致生理休眠。
- Abstract:
- Fritillaria meleagroides was taken as materials,the seed morphology was observed under scanning electron microscope.The viability of seeds was measured by TTC method.The moisture content of seeds was measured by oven weighing,the soaked seeds were used to test water permeability.The bioassay was applied for detecting the germination inhibitors.The changes of embryo and germination was observed by microscope.In order to understand the reason for seed dormancy and its particularity of Fritillaria meleagroides.The results showed that there were a lot of small stoma in the seed wing of the F.meleagroides under scanning electron microscope,and the seed coat was as thin as cicada′s wings.The water absorption of F.meleagroides seeds was more than 100% after being soaked for 2 hours.All of aqueous extract inhibited the germination and growth of Chinese cabbage.The change of embryo rate and the germination of seeds showed that the embryo development was not complete.Thus,there was no waterabsorbing barrier in F.meleagroides seed coat.F. meleagroides did not belong to the Fritillaria L.with mature embryo.Because the embryo was not fully developed,there was morphological dormancy,and the germination inhibitor leaded to physiological dormancy.
