- 以党参幼苗为试材,通过外源施加铜离子的方法,研究了铜胁迫下在基质中添加凹土对党参幼苗叶片生理学指标的影响。结果表明:铜离子胁迫降低了党参幼苗叶片的光下最大光化学效率(Fv′/Fm′)、光适应下实际光化学效率(Y(Ⅱ))、光化学猝灭系数(qP)、电子传递效率(ETR)和叶绿素含量;并升高了叶片可溶性糖含量、可溶性蛋白质含量、丙二醛含量和抗氧化酶(过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX))活性。非铜离子胁迫下,在基质中添加适量凹土能够提高党参幼苗叶片Fv′/Fm′和叶绿素含量的水平,但未对其它生理学指标产生显著性影响。在铜胁迫下,基质中含有适量凹土能明显减缓铜离子胁迫导致的党参幼苗叶片Fv′/Fm′、Y(II)、qP、ETR和叶绿素含量的降低以及叶片可溶性糖含量、可溶性蛋白质含量、丙二醛含量和3种抗氧化酶活性的升高。试验表明,在党参幼苗培养过程中,在基质中添加适量凹土可在一定程度上缓解铜胁迫对党参幼苗叶片的生理学压力。
- Abstract:
- Using the seedlings of Codonopsis pilosula (C.pilosula) as the plant material,and exogenous application of copper ion,the effect of attapulgite on the physiological responses of the leaves of C.pilosula seedlings to copper stress was studied.The results showed that copper stress decreased the levels of the maximal photochemical efficiency under light (Fv′/Fm′),photochemical efficiency under light adaptation(Y(II)),photochemical quenching coefficient (qP),electron transfer rate (ETR),and chlorophyll content of the leaves of C.pilosula seedlings.The copper stress also increased the contents of soluble sugar,soluble protein,malondialdehyde (MDA) and the activities of antioxidant enzymes (including peroxidase (POD),catalase (CAT),ascorbate peroxidase (APX))of the leaves of C.pilosula seedlings.Under condition without copper stress,the addition of certain amount of attapulgite into the growth medium enhanced the levels of Fv′/Fm′ and chlorophyll content of the leaves of C.pilosula seedlings,but did not significantly change other physiological indexes.Under the copper-stressed condition,the presence of certain amount of attapulgite in the growth medium significantly relieved the decreasing of Fv′/Fm′,Y(II),qP,ETR,and the contents of chlorophyll of the leaves and,simultaneously slowed down the increases of the contents of soluble sugar,soluble protein,MDA,and the activities of POD,CAT,APX of the leaves.These results indicated that during the seedling cultivation of C.pilosula,attapulgite in growth medium could be used as a beneficial component,to relieve the Cu-induced physiological stress.
