- 以采自河北农业大学苹果园的褪绿叶斑病毒(apple chlorotic leaf spot virus,ACLSV)样本为试材,采用实时荧光定量RT-PCR方法,研究了苹果不同组织部位、不同树体方位、不同生长发育时期和病毒相对含量之间的关系,以期为明确褪绿叶斑病毒在苹果植株中的时空分布规律提供参考依据。结果表明:从组织部位角度分析,在苹果的盛花期,ACLSV在花中的相对含量最高,芽中最低,ACLSV相对含量由高到低依次为:花、二年生枝皮组织、叶、一年生枝皮组织和芽。从树体方位角度分析,在苹果一年生枝皮组织中,ACLSV在4个方位上的病毒相对含量差异显著,位于南向的病毒相对含量最高,西向最低。从生长发育时期角度进行分析,2015年9月至2016年4月,苹果一年生枝皮组织中,病毒相对含量从9月中旬至10月中旬大幅升高,10月中旬达到这8个月中的最高峰,此后病毒相对含量开始下降,3月起呈上升趋势。

- The apple chlorotic leaf spot virus (ACLSV) samples collected from the apple orchard of Agricultural University of Hebei were used as materials,the real time fluorescence quantitative RT-PCR method was used to study the relationship between the different tissue type,the different direction of plant,the different stage of growth and development and the relative content of the virus,in order to provide references for defining the temporal and spatial distribution pattern of chlorotic leaf spot virus in apple plants.The results showed that from the view of tissue type,during full-blossom period,the relative concentration of ACLSV was the highest in flowers and the lowest in buds.The order from high to low was flowers,biennial branches bark,buds,annual branches bark and leaves.From the point of the direction of plant,using apple annual branches as material,the relative concentrations of ACLSV in four different directions were significantly different.The virus relative concentration in annual branch bark located in the south was the highest,the west was the lowest.From the view of the stage of growth and development,during the period of September 2015 to April 2016,using apple annual branches as material,the virus relative concentration increased from mid September to mid October.The relative concentration reached the peak in October and thereafter viral content began to decline and the trend showed upward in March.
