- 以废弃物蓝莓叶为原料,采用回流和超声2种方法,在单因素试验的基础上,采用正交实验优化了工艺条件,分别研究了溶剂浓度、提取温度、料液比、提取时间对蓝莓叶中的总黄酮得率的影响,确定了回流提取和超声提取的最佳条件,对提取产物总黄酮进行了显色稳定性试验和仪器精密度试验。结果表明:回流提取法的优化条件为料液比1∶5g·mL-1,提取温度100℃,提取时间90min,乙醇浓度70%,得率为7.24%;超声提取法的最佳提取条件为提取时间45min,温度为60℃,乙醇浓度70%,料液比1∶5g·mL-1,得率为7.18%。综合考虑,超声提取和回流提取优化后得率比较接近,二者相较而言,超声提取时间较短,对温度要求不高,可选择超声进行提取。

- Takingblueberryleavesastherawmaterial,totalflavonoidsfromblueberryleaveswasextractedbyultrasonicmethodandrefluxextraction,theeffectsofextractionconditionssuchassolventconcentration,extractiontemperature,ratioofliquid-solid,extractiontimeofextractionwereinvestigatedbyone-factortestsandorthogonaltest.Thecolorstabilitytestandinstrumentprecisiontestoftotalflavonoidswerecarriedout.Theresultsshowedthatoptimumextractionconditionsoftotalflavonoidsfromblueberryleavesbyrefluxmethodweresolid-liquidratio1∶5g·mL-1,extractiontime90minutes,70%ethanolconcentration,temperature100℃,theaverageextractionrateoftotalflavonoidswas7.24%;theoptimumextractionconditionsbyultrasonicmethodwereextractiontime45minutes,temperature60℃,70%ethanolconcentration,solid-liquidratio1∶5g·mL-1,theaverageextractionrateoftotalflavonoidswas7.18%.Consideredthatultrasonicextractionandrefluxextractionwererelativelyclosetoeachother,theultrasonicextractiontimewasshortandthetemperaturerequirementwasnothigh.Ultrasonicextractioncouldbeselected.
