- 以“津优1号”黄瓜子叶节为外植体试材,研究了外植体苗态、乙酰丁香酮(AS)浓度、脱菌次数、头孢霉素(Cef)浓度及生根培养基等因素对遗传转化的影响,以期为开展黄瓜的遗传转化研究提供参考依据。结果表明:使用子叶抱合类外植体诱导分化效果最好;在预培养基和共培养基中添加100 μmol?L-1AS有利于外植体分化;用无菌水冲洗3次可有效控制农杆菌污染;选择培养基中添加300 μmol?L-1Cef,生根培养基中添加400 μmol?L-1 Cef在抑制农杆菌的同时可有效促进根系生长。
- Abstract:
- ‘Jinyou No 1 ’ cucumber cotyledonary node explants was used as experimental material,the effects of seedling type,acetosyringone concentration (AS) in pre-culture medium and co-culture medium,washing times with sterile water,cephalosporin (Cef) concentration in select-culture medium and rooting medium on genetic transformation system were researched in order to supply reference for genetic transformation of cucumber.The results showed that various optimum parameters such as seedling type A(closed cotyledon),with 100 μmol?L-1 of AS,washed three times with sterile water,select-culture medium with 300 μmol?L-1 Cef,and rooting medium with 400 μmol?L-1 Cef were obtained with cotyledonary node explants.
