板栗针叶小爪螨体内Wolbachia的wsp基因序列测定与分析

- 以板栗针叶小爪螨为试虫,采用PCR扩增法,应用Wolbachia的wsp基因特异引物对我国部分板栗产区(北京密云南达峪村、山东泰安赵峪村与山东临沂柳条峪村)板栗园针叶小爪螨进行了检测,以期为通过分子生物学手段检疫鉴定针叶小爪螨提供参考依据。结果表明:在北京密云南达峪村与山东泰安赵峪村板栗园针叶小爪螨体内扩增出593 bp Wolbachia的wsp基因;在山东临沂柳条峪村板栗园针叶小爪螨体内未检测到Wolbachia特征基因片段。利用所测序列和其它已发表的序列建立系统树,发现针叶小爪螨体内Wolbachia属于B大组的Epo小组。
- Abstract:
- Oligonychus ununguis was used as test insect.Specific primers derived from the wsp gene sequence were used to amplify DNA of Wolbachia from part of chestnut orchards(Miyun Beijing,Tai′an Shandong and Linyi Shandong chestnut orchards) of Oligonychus ununguis in China by polymerase chain reaction (PCR),in order to supply reference for detecting Oligonychus ununguis by molecular biological method.The results showed that the 596 bp products of wsp gene were observed after amplification of extracts from chestnut orchards (Nandayu village of Beijing and Zhaoyu village of Shandong Province) of Oligonychus ununguis,but in other chestnut orchards (Liutiaoyu village of Shandong Province),this gene was not found.By analyzing the wsp gene sequences of Wolbachia in Oligonychus ununguis by constituting the phylogenetical tree,it was found that they were identical with the wsp gene sequence of some strains of Epo group in B supergroup of Wolbachia.
