- 以种植12年的设施温室土壤线虫群落为研究对象,采用土壤线虫18S rDNA克隆文库方法,研究了土壤线虫物种多样性指数、植物寄生线虫成熟度以及自由生活线虫成熟度等土壤线虫群落结构特征,以期揭示土壤健康程度。结果表明:设施蔬菜土壤线虫以食细菌线虫为优势类群,占59.4%;植物寄生线虫比例次之,占34.9%。在属水平上,以根结线虫为优势类群,比例为31.5%,其次为拟小杆属线虫,比例为28.8%。设施温室土壤线虫的成熟度指数和植物寄生线虫成熟度指数分别为1.86和1.11。总体来看,设施温室土壤中植物寄生线虫比例较高,这反映出设施温室土壤受到人为干扰较大,且线虫营养类群结构失调,土壤健康状况不良。
- Abstract:
- In order to analyze the characters of soil nematode community in greenhouse 12 years history,and to assess the status of soil health in greenhouse,the community of soil nematode was investigated to reveal the diversity of soil nematode community,plant parasite index (PPI) and mature index (MI) by soil nematode 18S rDNA clone library.The results showed that the dominant soil nematode group of 12 years greenhouse were bacterial-feeding nematodes with percents of 59.4%,followed by plant parasitic nematode with percent of 34.9%.At genus level,Meloidogyne was the predominant nematode groups represented 31.5%,followed by Rhabditoides with 28.8%.Moreover,the PPI and MI of soil nematode community in 12 years greenhouse were 1.11 and 1.86,respectively.In a conclusion,plant parasitic nematode possessed high percentage in soil nematode community,suggesting that soil was disturbed by farm operation and structure of soil nematode community disorder.On the whole,health of soil in greenhouse was abnormal.
