- 以穿心莲为试材,采用薄层色谱法(TLC)对穿心莲内酯与脱水穿心莲内酯进行定性鉴别,以三氯甲烷-乙酸乙酯-甲醇(4∶3∶0.4)为展开剂,以2%的3,5-二硝基苯甲酸乙醇溶液-2 mol?L-1氢氧化钾溶液(1∶1)混合溶液为显色剂;并采用高效液相色谱法(HPLC)测定穿心莲内酯、脱水穿心莲内酯的含量,使用Hypersil BDS C18(4.6 mm×250 mm,5 μm)为色谱柱,以甲醇-水(65∶35)为流动相,柱温30 ℃,穿心莲内酯检测波长为225 nm,脱水穿心莲内酯检测波长为254 nm。结果表明:建立的薄层色谱法能检出穿心莲内酯与脱水穿心莲内酯,HPLC法能够准确测定穿心莲中穿心莲内酯与脱水穿心莲内酯的含量。使用薄层色谱鉴别及高效液相色谱法快速、准确,重复性好,可用于穿心莲药材的质量控制。
- Abstract:
- With Andrographis paniculata as test material,andrographolide and dehydroandrographolide were identified by TLC,using chloroform-ethyl acetate-methanol(4∶3∶0.4) as the mobile phase and 2% 3,5-diniotrbenzoie acid ethanol-2 mol?L-1 potassium hydroxide(1∶1) as the chromogenic agent;and the content of andrographolide and dehydroandrographolide was detected by HPLC.The separation was carried out on a Hypersil BDS C18(4.6 mm×250 mm,5 μm) column with the mobile phase of methanol-water(65∶35);the column temperature was at 30 ℃;the detection wavelength of andrographolide and dehydroandrographolide were 225 nm and 254 nm,respectively.The results showed that andrographolide and dehydroandrographolide could be identified by TLC,the content of andrographolide and dehydroandrographolide in Andrographis paniculata could be accurately determined by HPLC.The established methods of TLC and HPLC might be a reliable and specific method with good reproducibility,which could be used as quality control method of Andrographis paniculata.
