- 以33份暗褐网柄牛肝菌样品为试材,采用SSR标记技术,对其遗传多样性进行研究,以期为暗褐网柄牛肝菌商业菌株鉴定奠定基础。结果表明:15个SSR标记中有5个标记在供试样品中表现出多态性,每个标记检测到2~6个等位基因,平均3.20个,Shannon′s 信息指数平均值0.76,多态性信息含量平均值0.43。聚类分析结果显示,在相似系数为0.99时,供试材料可被分为11个类群,对应11种多位点分子表型,与样品的来源有较好的对应关系。根据多位点分子表型构建了供试材料的SSR指纹图谱。
- Abstract:
- In this study,the genetic diversity of 33 Phlebopus portentosus materials was analyzed by using SSR markers in order to lay the foundation of commercial strains indentification of Phlebopus protentaosus.The results showed that 5 out of 15 SSR markers represented polymorphism in testing samples,2-6 alleles were found in each marker and the mean value was 3.20.The average values of Shannon′s information index and polymorphism information content were 0.76 and 0.43 respectively.The testing samples were clustered into 11 groups at the genetic similarity 0.99 on a UPGMA dendrogram,which corresponded to 11 multiple loci molecular phenotypes and the cloning origin of the samples.The codes of SSR fingerprinting were constructed based on these multiple loci molecular phenotypes.
