葡萄柚成年态茎段外植体的消毒与抗褐化

- 以葡萄柚不同品种的成年态茎段为外植体,研究了不同茎段长度、品种、消毒方式及培养基对葡萄柚成年态茎段消毒以及防褐化的影响。结果表明:2 cm大小茎段消毒效果最好,污染率最低,萌芽率最高。不同品种的污染率和发芽率存在差异,“里约红”污染率最低,“星路比”污染率较高,但萌芽率最高。用75%酒精处理20 s,5% NaClO溶液处理20 min,放置48 h后,再用5% NaClO溶液处理20 min,污染率最低达21.3%,为最佳消毒方法。NaClO二次消毒后接种于WPM+100 mg?L-1半胱氨酸培养基上可有效控制褐化,污染率最低为15.6%,萌芽率最高为83.1%。
- Abstract:
- Stem segments of grapefruit were used as explants,the effects of different stem lengths,seedling varieties,disinfection methods and media types on disinfection result and browning prevention were studied.The results showed that 2 cm of stem section was the best disinfection effect.Difference of contamination and sprout rate existed among cultivars.‘Rio Red’ pollution rate was the lowest,the ‘Star Ruby’ was higher,but it had the highest germination rate.After treated by 75% ethanol for 20 seconds and followed by 5% hypochlorous sodium for 20 minutes,the materials were kept in culture room for 48 hours,then the material was treated by 5% hypochlorous sodium for 20 minutes again.This was the best way to disinfect with the minimum pollution rate 21.3%.With sodium hypochlorite secondary sterilization after vaccination in WPM+100 mg?L-1 cysteine medium,it could effectively control the degree of browning and get minimum pollution rate,only 15.6%,the highest germination rate was 83.1%.
