- 以甘菊为试材,采用基因克隆及生物信息学方法,克隆了甘菊ClCOL〖STBX〗5a基因并对其进行生物信息学分析及表达分析。结果表明:该基因共编码347个氨基酸,编码序列中含有2个保守的B-Box结构域和1个CCT结构域,属于CO家族的I类基因。NCBI Blastp分析与进化树分析均显示,该基因编码产物与拟南芥CO家族中AtCOL5相似性最高,故将该基因命名为ClCOL5〖STBX〗a。实时定量分析显示,ClCOL〖STBX〗5a为组成型表达,且在叶中表达量最高。在花芽膨大的过程中,ClCOL〖STBX〗5a基因的表达逐渐升高,并在露色时达到最大值,之后逐渐降低,表明ClCOL〖STBX〗5a在甘菊开花过程中可能发挥着重要功能。
- Abstract:
- Taking Chrysanthemum as test material,the ClCOL〖STBX〗5a gene was cloned and analyzed.The results showed that this gene encoded a protein of 347 amino acids containing two B-Box domains and one CCT domain,which indicated that it belonged to the CO family group I.Both the results of NCBI Blastp and phylogenetic tree based on CO proteins showed it was mostly closed to AtCOL〖STBX〗5 gene than other member of the family,so it was named as ClCOL〖STBX〗5a.Relative real time PCR analysis showed that ClCOL〖STBX〗5a expressed in all the tested tissues,with the highest level in leaf.In the process of flowering,the expression of ClCOL〖STBX〗5a gene was gradually increased,and reached the maximum level at pigmented stage,and then decreased gradually,which indicated that ClCOL〖STBX〗5a might play an important role in the development of flower in Chrysanthemum.
