- 以苹果“平邑甜茶”(Malus hupehensis var.pingyiensis)为微嫁接砧木,“寒富”(Malus domestica cv.‘Hanfu’)为接穗,研究了附加不同浓度6-BA的培养基、接穗不同生理状态、接穗具有叶片数量、不同接穗/砧木组合对嫁接成活率和嫁接口愈合的影响。结果表明:最适于微嫁接的培养基为MS+IAA 0.3 mg/L+BA 1.0 mg/L,选用继代培养40 d的接穗和砧木,并且接穗部分具有2片真叶时的嫁接成活率最高;用“平邑甜茶”作为砧木的嫁接成活率高(81.3%),嫁接口愈合最好。该研究为下一步的转基因sRNA信号在砧木与接穗间的传递运输研究奠定基础。
- Abstract:
- Taking ‘Pingyitiancha’ (Malus hupehensis var.pingyiensis) as rootstock and ‘Hanfu’(Malus domestica cv.‘Hanfu’) as scion,the effect of different concentrations of 6-BA,ages of scion,leaves number of scion,kinds of scion/rootstock on the grafting survival rate was researched.The results showed that MS+IAA 0.3 mg/L+6-BA 1.0 mg/L,rootstocks and scions were cultured for 40 days,with 2 leaves on the scion and ‘Pingyitiancha’ was used as the rootstock,the survival rate of micrografting was 81.3%.Those were the foundation of transferring sRNA from rootstocks to scions.
