- 以西洋参种胚诱导愈伤组织上的体细胞胚(胚状体)为试材,采用离体培养方法,研究胚状体自身发育阶段及用于成熟、萌发生长的培养基的组成因子对其成熟萌发的影响,以期确定最佳的胚状体成熟培养基和萌发培养基。结果表明:发育阶段为鱼雷形的胚状体,是进行成熟培养的最佳材料;胚状体成熟的最佳培养基是0.5M+0.4 mg/L 6-BA+2 mg/L NAA+5 mg/L GA3+1%AC;在此培养基上胚状体生长变大,子叶生长展开,胚芽、胚根肉眼可见;成熟培养基中植物生长调节物质在活性炭作用下缓慢释放并作用于胚状体,达到形成大量正常、健壮、同步胚状体的目的;胚状体萌发的最佳培养基是0.5MA+0.4 mg/L 6-BA+2 mg/L NAA+5 mg/L GA3;在此培养基上胚状体萌发生长,胚芽伸长真叶展开。
- Abstract:
- Taking Panax quinauefolium L.embryo tissue of somatic embryo callus (embryoid) as test materials,using the method of in vitro culture,the effect of somatic embryo formand composition of medium for its mature and germination were investigated,in order to determine the best grow mature embryoid culture medium and culture medium.The results showed that the torpedo of somatic embryos was the best form transferred for mature culture;the best mature medium 0.5M+0.4 mg/L 6-BA+2 mg/L NAA+5 mg/L GA3+1% AC,in which somatic embryos grew bigger,cotyledon expanded,germ and radicle grew visible to the naked eye;plant growth regulating substances under the effect of activated carbon released slow,purposed to form a large number of normal,robust,synchronous somatic embryo were achieved;the best germination medium was 0.5MA+0.4 mg/L 6-BA+2 mg/L NAA+5 mg/L GA3,in which somatic embryos germinated,its plumule elongated and the leaf expanded.
