- 以杜梨根瘤为试材,采用分子生物学方法、牛津杯法和指示植物接种法,研究了杜梨根癌病原菌的质粒类型及枯草芽孢杆菌对杜梨根癌病原菌的影响。结果表明:从1年生杜梨根部冠瘿瘤上分离、纯化、培养菌株,得到4株与根癌病原菌相似菌株,对之进行PCR扩增,此4株菌株均获得特异性目的条带,鉴定为根癌土壤杆菌胭脂碱类型。将病原菌接种指示植物向日葵幼苗,4株菌株均有致病性。经室内离体试验,供试的9株枯草芽孢杆菌对病原菌表现不同程度的抑菌效果;接种指示植物试验中,菌株9076和9161过滤发酵液能够完全抑制冠瘿瘤的生长。
- Abstract:
- The plants of Pyrus betulaefolia Bge that affected by crown gall pathogen were applied in this experiment.Molecular biology methods were used to identify the plasmid of pathogen of P.betulaefolia Bge crown gall.The results showed that the bacteria sampled from the crown gall that grew on the roots of the one-year-old P.betulaefolia plants were cultured and purified.Four bacterium strains similar to Agrobacterium were obtained.PCR assay showed that four bacterium strains had specific band and could be classified to the type of nopaline.These strains caused crown gall after inoculated into the stems of the sunflower seedlings as index plants.The oxford cup method and the index plants inoculated with both Bacillus subtilis and the bacteria of crown gall were used to study the effect of Bacillus subtilis against the pathogen of P.betulaefolia Bge.The results showed that nine Bacillus subtilis strains showed inhibition to the Agrobacterium strains in in vitro test.Inoculating of the supernatant of Bacillus subtilis strains 9076 and 9161 showed completely resistance to the growth of the crown gall in the index plants..
