- 以铁皮石斛T4种群原球茎为试材,研究了原球茎团块直径、接种密度、不同激素配合及继代周期对原球茎的增殖和长势的影响,以期构建铁皮石斛原球茎的高效增殖体系。结果表明:在铁皮石斛原球茎的增殖过程中具有明显的群体效应,适当增大原球茎团块直径和接种密度有利于提高增殖率;不同浓度的激动素(KT)、6-苄基腺嘌呤(6-BA)与萘乙酸(NAA)配合试验中,NAA 0.02 mg/L+KT 0.5 mg/L能更有效地促进原球茎的增殖和生长;铁皮石斛原球茎增殖培养的适宜继代周期是50 d。
- Abstract:
- Taking the protocorm of T4 population as trial material,the effect of mass diameter,inoculation density of protocorm,hormone combinations,subculture cycle on proliferation and growth of protocorm were researched for establishing a highly efficient proliferation system of protocorm of Dendrobium officinale Kimura et Migo.The results showed that the high community effect could be observed during protocorm proliferation culture,so the appropriate increase of mass diameter and inoculation density of protocorm was of benefit to protocorm proliferation.Experiment was done with different concentration combinations of KT added with NAA or 6-BA added with NAA,which showed that NAA 0.02 mg/L+KT 0.5 mg/L was the best hormone combinations for the proliferation and growth of protocorm.50 d was the suitable subculture cycle for protocorm proliferation culture of Dendrobium officinale Kimura et Migo.
