- 以木奈(Prunus salicina Lindl.var.cordata J.Y.Zhangetal)5个不同生长发育时期叶片为试材,根据已构建木奈5K.tif,JZ的5个不同生长发育时期叶片的均一化全长cDNA文库,分离得到了一个羟脂酰CoA脱水酶(HCD)基因,命名为PsHCD 。对PsHCD基因生物信息进行分析,同时利用Real-time PCR检测PsHCD基因在叶片中5个不同生长发育时期的表达量。结果表明:该基因的cDNA开放阅读框编码区为第322个核苷酸到第987个核苷酸,编码221个氨基酸残基,5′UTR长度为321 bp,3′UTR长度为156 bp。利用TMHMM server软件分析,该基因编码的蛋白质有4个跨膜区域,PSORT Ⅱ Prediction软件分析推测,亚细胞定位于内质网、微体和溶酶体中。实时荧光定量PCR结果表明,PsHCD的表达量随着叶片生长逐渐升高,在展开叶时达到最高峰,在幼叶到成熟叶时处于一种稳定水平,随后降低,老叶表达量最低。
- Abstract:
- Taking five different growth period of Prunus salicina’s leaves as material,a clone named PsHCD was separated from cDNA library prepared of five different growth and development periods of leaf in the Prunus salicina.The biological information of PsHCD was analyzed and gene expression levels in five different growth periods of Prunus salicina’s leaves were tested using Real-time PCR.The results showed that the gene cDNA open reading frame coding regions for the 322thnucleotide to 987th,encoding a 221 amino acid proteins,with 321 bp in the 5’UTR and 156 bp in the 3’UTR.Using TMHMM server software to analysis,the gene encoding the protein had four transmembrane domains.PsHCD was localized in endoplasmic reticulum,microbodies and lysosomes by PSORT Ⅱ Prediction software.The results of Real-time PCR showed that the PsHCD gene was gradually increasing with development of leaves,reached to maximum at stage of unfold leaves,was stable during spire to climax leaves,then decreased,reached to minimum at stage of old leaves.
