- 以苦豆子为试材,采用组织培养的方法,研究了外植体种类、激素水平、水解酪蛋白浓度等不同因子对愈伤组织生长状态的影响,同时对愈伤组织进行了去褐化处理。结果表明:苦豆子子叶为诱导愈伤组织的最佳外植体;最佳诱导培养基为MS+2,4-D 0.5 mg/L+NAA 0.5 mg/L+6-BA 1.0 mg/L;最佳继代培养基为MS+2,4-D 1.0 mg/L+6-BA 4.0 mg/L+NAA 1.0 mg/L+CH 1 000 mg/L;添加3%蔗糖的条件下快速继代是较好的去褐化措施。
- Abstract:
- Taking Sophora alopecuroides as material,the effect of explants type,hormone levels,casein hydrolysate on the growth of callus were measured by tissue culture method.Meanwhile many measures were carried out to prevent browning.The results showed that the cotyledon was the optimum explants of callus induction,the best medium of callus induction was MS+2,4-D 0.5 mg/L+NAA 0.5 mg/L+6-BA 1.0 mg/L;and the best subculture medium was MS+2,4-D 1.0 mg/L+6-BA 4.0 mg/L+NAA 1.0 mg/L+CH 1 000 mg/L;the best proposal was frequent subculture with 3% sucrose.
