- 以万寿菊‘QU2-2’为试材,从取材时间、根的长度、预处理方法、解离时间、染色时间、压片工具6个方面对根尖染色体制片方法进行了筛选和优化。结果表明:万寿菊根尖染色体观测采用室内催根培养,于上午9:14,剪取根长1.0 cm的根尖,利用低温(0~4℃)水浴进行预处理24 h、1.0 mol/L盐酸解离7 min、染色25 min、结合带橡皮头铅笔压片,可以获得高质量的有丝分裂前中期临时片。
- Abstract:
- Taking the root-tip of Tagetes erecta L.‘QU2-2’ as the experimental material,the material taking time,length of the root,material pretreatment method,dissociating time,dyeing time and squashing tools were screened and optimized.The results showed that using indoor root culture,observed the high-quality images of the chromosomes of root-tips at 9:14,1.0 cm of the root length,the cool water pretreatment (0~4℃),1.0 mol/L HCl for dissociation in 7 minutes,dyeing in 25 minutes,squashing the slides with a rubber head pencil,could observe clear mitotic prometaphase.
