- 以“青芋1号”和“神芋”菊芋品种为试材,通过自然状态的低温锻炼,测定了2个菊芋品种块茎中总糖、还原糖、可溶性糖及淀粉含量的变化;并利用扫描透射电镜观察了低温锻炼下的块茎细胞超微结构。结果表明:抗寒性品种“青芋1号”随温度降低,总糖含量升高,还原糖含量升高,可溶性糖含量增加,淀粉含量降低;反之,非抗寒性品种“神芋”总糖含量降低,还原糖升高幅度小,可溶性糖含量降低,淀粉含量降低幅度大。细胞超微结构发现,抗寒性品种“青芋1号”在-20℃时才表现出液泡破碎,线粒体数量增多,变形及解体;非抗寒性品种“神芋”在5℃时就出现类似现象,这与植物在遭受寒冷环境条件细胞适应性变化相一致。
- Abstract:
- Using ‘Qingyu No.1’ and ‘Shengyu’ as materials,the content of total sugar,reducing sugar,soluble sugar and starch of two varieties Jerusalem artichoke tuber under the nature low temperture stress were determined.Meanwhile,tuber cells ultrastructure under cold stress by scanning transmission electron microscopy were also observed.The results showed that ‘Qingyu No.1’ had total sugar increasing,reducing sugar increasing,soluble sugar increasing,starch reducing trend.Otherwise,‘Shenyu’ had total sugar reducing,reducing sugar small increasing,soluble sugar reducing,starch more reducing trend by statistical analysis.The results of cells ultrastructure observation showed that cold resistance tuber ‘Qingyu No.1’ was vacuole crushing,the number of mitochondria increasing,deformation,disintegration under -20℃,but showed by non-cold resistance tuber ‘Shenyu’ under 5℃,this order was identical with plants cytop cell defense phenomenon cope with cold environment condition.
