- 以沙棘品种“实优1号”的1~2 a生带休眠芽茎段(腋芽未萌发)为外植体,采用不同培养方式、不同类型基本培养基和不同激素浓度组合对沙棘组织培养过程中的影响进行系统的研究。结果表明:茎段分化培养采用1/3MS+6-BA 0.5 mg/L+IAA 0.3 mg/L,诱导率可达85%以上;最佳增殖和诱导愈伤培养基为1/3MS+6-BA 0.75 mg/L+IAA 0.5 mg/L,愈伤诱导率在90%以上,增殖系数达到4~6;最佳生根培养基均为1/4MS+NAA 0.3 mg/L+IBA 0.5 mg/L,平均生根率85%以上。接种后进行暗培养7 d,向培养基中添加6.5 g/L琼脂,1.5 g/L活性碳,可有效防治褐化。
- Abstract:
- Through the ‘Real gifted one’ of Hippophae rhamnoides varieties with 1~2 years old and dormant buds stems (axillary bud of unerupted) as explants,different culture,different types of basic culture medium and different concentration of hormone combination on Hippophae rhamnoides tissue culture influence in the process of systematic were studied.The results showed that the stem segment culture medium for differentiation was 1/3MS+6-BA 0.5 mg/L+IAA 0.3 mg/L;and the induction rate could reach more than 85%;the best of proliferation and induction of callus culture medium was 1/3MS+6-BA 0.75 mg/L+IAA 0.5 mg/L;callus induction rate in 90% above,and proliferation coefficient reached 4~6;the optimum rooting medium was 1/4MS+NAA 0.3 mg/L+IBA 0.5 mg/L;average rooting rate above 85%.After inoculation dark culture was added into the culture medium 7 d,6.5 g/L agar,1.5 g/L activated carbon,more training method could effectively prevent the browning.
