Abstract:
A total of 33 wild Bletilla striata Rchb.f.from six different province were studied with 13 pairs of SRAP combining primers,clustering analysis and fingerprint construction of 33 germplasm resources of Bletilla striata Rchb.f.was carried out,in order to provide reference of plant resourceses protection and mixed falsify for the appraisal.The results showed that 13 SRAP combining primers produced 182 bands,included 152 polymorphic bands,with a polymorphism ratio of 83.52%.The average Shannon′s information index and the average gene diversity index were 0.484 6 and 0.324 3.Based on UPGMA,six populations gathered in two clusters.The cluster Ⅰ included three populations.Three populations were respectively taken from Guangdong province,Guangxi province and Hunan province.The another three populations which came from Sichuan province,Guizhou province and Yunan province belonged to the cluster Ⅱ.Mantel test revealed significant positive correlation between genetic distances and geographic distance (r=0.624 6;P<0.001).The DNA fingerprinted of 33 germplasm resources of Bletilla striata Rchb.f.were constructed by with four core SRAP combining primers.
