Abstract:
Taking middle-layer scales of the dual-purpose ornamental and edible cultivar ‘Yandan No.1’ (Lilium pumilum DC.) as test material,a three-factor experimental design-seven sucrose concentrations (30-90 g·L-1),light and dark two conditions.Four sampling time points was employed,and three-way ANOVA together with simple-effects analysis was used to investigate the effects of sucrose concentration and light-dark conditions on bulblet weight gain,enlargement,differentiation,rooting and leaf emergence during tissue-culture rapid propagation,in order to provide a reference for the large-scale production of ‘Yandan No.1’.The results showed that sucrose concentration,light/dark treatment,and culture time had significant or highly significant effects on all growth indicators,and interactions were observed for most indicators.Light conditions generally favored adventitious bud differentiation,root formation,and leaf emergence,whereas dark conditions promoted fresh weight or diameter increase in some medium-and high-sucrose treatments; however,these promotive effects were influenced by sucrose concentration and culture time.Under light conditions,40 g·L-1 sucrose treatment (A2B1) showed relatively stable differentiation,60 g·L-1 sucrose treatment (A4B4) showed better rooting performance,and 50 g·L-1 sucrose treatment (A3B1) showed better comprehensive performance in fresh weight,enlargement,and stability.When leaf emergence at 60 days was used as the criterion,A2B1 performed better.Considering weight gain,enlargement,differentiation,rooting,leaf emergence,and coefficient of variation,50 g·L-1 sucrose under light culture could be recommended as a suitable combination for high-quality bulblet production of ‘Yandan No.1’.
