Abstract:
Taking 20113 and 21014 Phlebopus portentosus strains as the test materials,the effects of temperature and pH on the production of starch hydrolases were investigated with the method of the Congo red-starch test and Lugol′s iodine-starch test,in order to provide reference for the development of an efficient starch utilization strategy for Phlebopus portentosus.The results showed that in the Congo red-starch test,with increasing temperature,the mycelial growth rate and starch hydrolases activity gradually increased and reached a maximum at 28 ℃,followed by a gradual decrease.In the Lugol′s iodine-starch test,with increasing temperature,the mycelial growth rate and starch hydrolases activity gradually increased and reached their maximum at 30 ℃ and 28 ℃,respectively,followed by a gradual decrease.With increasing initial pH,the starch hydrolases activity gradually increased and reached its maximum at pH 6.5,with no significant change between pH 7.0 and 8.0.As pH increased,the mycelial growth rate of the 21014 strain gradually decreased,while the mycelial growth rate of the 20113 strain did not show significant changes.In conclusion,the optimal temperature for the production of starch hydrolases by Phlebopus portentosus was 30 ℃,and the optimal initial pH was 6.5.
