Abstract:
Taking the seedlings of Trollius chinensis Bunge.in vitro as test materials,the effects of explant type,basic medium and exogenous hormone on callus induction and adventitious bud regeneration were studied by tissue culture method in order to establish its rapid propagation system.The results showed that the optimal explants were cotyledons from seedlings of Trollius chinensis Bunge.MS medium was the best medium for callus induction.Medium plus 2.0 mg?L-1 6-BA and 1.0 mg?L-1 2,4-D was advantageous to callus induction,and callus rate was 72.00%.Maltose 30 g?L-1 as carbon source was better than sucrose.The medium adding activated charcoal 75 mg?L-1 could prevent brown efficiently.The differentiation medium adding 0.5 mg?L-1 NAA and 1.0 mg?L-1 KT,could improve the differentiation rate of callus.
